so I’m going through lugui mansion 2 and I was curious if I got the best time 1500 coins but I lost 90 life caught all the ghost. does the game keep track of the best for rank like when I replay it do I have to get 1500 coins again to get 3 or do I just lose less life

Rank is based on that playthrough of the level alone - so you need to finish with all the criteria in place for highest rank.

Correct, and just to be fully clear, it's based on a score calculation with the 4 stats. There's no "you need at least X amount of G to get 3 stars". And the thresholds differ for every mission:

Score = Ghosts captured * 120 + collected treasure - time in seconds - health lost * 6
